Topical Diclofenac-Induced Hepatotoxicity

چکیده

The hepatotoxic potential of diclofenac, a commonly used non-steroidal antiinflammatory agent, is well established in literature. However, cases diclofenacinduced liver disease have occurred almost exclusively with the oral formulation this medication. We report case an elderly man Paget’s and osteoarthritis who developed acute hepatotoxicity, as evidenced by laboratory diagnostics, four months after use topical diclofenac 1% gel. Once gel was discontinued, repeat blood work three weeks discharge demonstrated return function tests to baseline. Given temporal relationship between initiation escalation changes tests, likelihood diclofenac-induced injury deemed possible using wellrecognized causality assessment tool. Further research on topically administered nonsteroidal anti-inflammatory agents needed identify monitoring intervals for early detection avoidance adverse effects patients diclofenac.

Diclofenac-induced hepatotoxicity.

Two patients developed clinical, biochemical and histopathological signs of a liver hypersensitivity reaction following treatment with diclofenac. Hepatic side effects of this drug are very rare. The relevant literature is reviewed.

Involvement of Lysosomal Labilisation and Lysosomal/mitochondrial Cross-Talk in Diclofenac Induced Hepatotoxicity

In this research, we investigated the cytotoxic mechanisms of one of the widely used pharmaceuticals that are regularly associated with the adverse effects on the liver, sometimes leading to acute liver failure, diclofenac. Diclofenac liver cytotoxicity was associated with reactive oxygen species (ROS) formation and lipid peroxidation which were inhibited by antioxidants and ROS scavengers, fer...

Topical Delivery of Diclofenac using Microemulsion Systems

The purpose of this study was to investigate microemulsions as delivery systems for diclofenac sodium (DS). Microemulsion systems were composed of: soybean oil, nonionic surfactants (Brij 58 and Span 80), and different alcohols: ethanol, and 1-buthanol as cosurfactant. The optimum surfactant: cosurfactant (S:CoS) weight ratios and microemulsion areas were detected by the aid of ternary phase di...
